O NAD(P)H quinona oxidoreductase-1 (hNQO1) humano é um importante biomarcador relacionado com o cancro, que mostra uma significativa sobreexpressão em células malignas. O desenvolvimento de um método eficaz para detectar a actividade de NQO1 com alta sensibilidade e selectividade nos tumores tem um grande potencial para o diagnóstico, tratamento e gestão do cancro. No presente estudo, relatamos uma nova sonda fluorescente à base de dicanoisophorona (DCP) capaz de monitorizar a actividade de hNQO1 in vitro e in vivo, tanto no modelo ratiométrico como no modelo "turn-on". A NQ-DCP foi preparada conjugando a fluoroproteína dicanoisoforona com ácido propiónico quinona activável hNQO1 (QPA), que permanece não fluorescente até à activação por hNQO1 específico do tumor. O NQ-DCP apresentava um grande Stokes shift (145 nm), excelente biocompatibilidade, permeabilidade celular, e selectividade em relação ao hNQO1, o que permitiu diferenciar as células cancerosas das células saudáveis. Utilizámos com sucesso o NQ-DCP para monitorizar a actividade endógena não invasiva do hNQO1 em células tumorais do cérebro in vitro e em tumores xenografados desenvolvidos em ratos nus.
